Under Fluorescence Microscope

Fungal spores are the main reproductive organ of fungi. They germinate under suitable conditions, develop hyphae, and multiply. Spores are the main media for spreading fungal pathogen infection to animals, plants, and humans. 

Direct checking under the microscope is a primary diagnosis method of fungal examination, which is on purpose to find mycelium and spores. Ordinary Gram staining is not specific, but fungi show character under fluorescence excitation wavelength at 380nm.

Gram staining is a method used to identify bacteria. This staining method uses the different biochemical properties of bacterial cell walls to divide bacteria into Gram Positive and Gram Negative categories. It was originally used to identify the relationship between pneumococcus and Klebsiella pneumoniae. It was later promoted as one of the important characteristics for identifying bacterial species and has a wide range of uses in the clinical diagnosis and treatment of diseases caused by bacterial infections.

It is seen bright blue fluorescence signal is excited by the UV light excitation wavelength 330nm~380nm. Compared to the gram-stained fungus it is easier to be found and gets higher clarity.

Limitations of this assay

The signal is not auto-fluorescence from the fungus but stained by the mixed fluorescence reagent.

Fluorescence observation is only effective if the user uses fluorescent dyes or if there is a local company that produces or supplies this mixed reagent.
